Permeabilization Buffer for ICC and IHC

Permeabilization Buffer is used for permeabilization of cells and tissue sections to enhance interaction of primary antibodies with cytoplasmic and nuclear target proteins in fixed samples by using immunocytochemistry (ICC) and immunohistochemistry (IHC) applications. The buffer can be used on specimens fixed with different fixatives including, but not limited to, buffered formalin, formaldehyde, alcohols, acetone and chloroform.

Typical application of the Cell & Tissue Blocking Buffer is as follows:

1.         Pre-incubate fixed samples with Permeabilization Buffer (30-100 uL per tissue section or cytosmear) for 10-30 min at room temperature;

2.         Discard Permeabilization Buffer and add Blocking Buffer, SF40011, ( 30-100 uL per tissue section or cytosmear) for 10-30 min at room temperature;

3.         Discard Blocking Buffer (it is not necessary to rinse samples with PBS, TBS and other wash buffers after discarding the Blocking Buffer);

4.         Add primary antibodies diluted with Neuromics Antibody Dilution Buffer (SF40010) and incubate specimens for a specified period of time.
